A proportional but slower NMDA potentiation follows AMPA potentiation in LTP (Turrigiano, NN, 2004)

• In the culture cortical neuron LTP, AMPA-mEPSC increases rapidly (<30 min). NMDA-mEPSC increases later (2 hours).
  • LTP induction protocol: 15 min. 37deg in NaCl 126 mM; KCl 5.5 mM; MgSO4 0.4 mM; NaH2PO4 1 mM; NaHCO3 25 mM; CaCl2 2 mM; dextrose 14 mM; glycine 0.2 mM; bicuculline 0.01 mM
• EPSC measured in paired whole-cell recordings from thick-tufted L5 neurons in 300 m slices of rat (P14–18) visual cortex showed similar results
  • LTP protocol: current-clamp mode by pairing 200-ms pre- and postsynaptic current injections (0.9–1.2 nA) 30 times at 0.1–0.2 Hz).
